Overview
In this I will show you how to unlock every character in the game. I am not including tainted characters as they are all unlocked by reaching home with that character. I will include Jacob/Esau and Bethany from Repentance though. Some are Easy some are hard to unlock but they are all worth it. Plus if you want to 100% beat the game you need to beat every boss with every character anyway.

All of the characters in Binding of Isaac unlock an item when they beat a Key boss. For example When you beat Satan as Isaac you unlock an item called Mum’s knife. Some items are better than others but this guide isn’t about items, it’s about the characters! You start with Isaac as the first character as he is the main one in the story.
To unlock Magdalene you need to have at least 7 red heart containers in 1 run. The best way to do this is by always beating the boss on that floor for the chance of a HP up item, this is so you have a chance rather than using a rune ( Or any other item ) to go to the next floor. Another tip is to give coins to the Coin Beggars because they have a chance of giving a HP up item.
To unlock Cain you need to have 55 coins in 1 run. A tip for this one is DON’T spend coins in the shop or use your coins in any other way. You might have to get a good seed for this one but Cain is worth the struggle.
To unlock Judas you need to defeat Satan. To reach Satan you must go in the trapdoor after defeating It lives or the trapdoor in the devil room if you have not unlocked it lives. A good tip for this is to take a devil deal earlier in the run so you don’t have the chance of getting an angel room instead.
To unlock ??? ( also known as blue baby ) You need to defeat Mum’s heart 10 times. This is pretty simple you are just going to have to be patient and get good runs until you do this.
To unlock Eve you need to avoid picking up any hearts for two floors in a row. A good tip is to play as Magdalene when doing this as she has 4 heart containers and has yum heart which gives you a heart. Yum heart doesn’t count as picking up hearts so she is a good character for this.
To unlock Samson you need to avoid taking damage for two floors in a row. This might be tricky if you are really new to the game and have not played that much but eventually you will unlock him. There isn’t really tips for this unlock.
To unlock Azazel you need to make 3 deals with the Devil in one game. Like before Magdalene is a good character for this because she has more hearts to make more deals. Azazel is a good character and I think this is a character you should unlock quickly.
To unlock Lazarus you need to have 4 or more soul or black hearts in a single run. A good tip is to blow up tinted rocks for a chance of soul hearts. Another tip is to go into spike door rooms if you don’t have any soul hearts yet because these rooms can contain soul / black hearts.
To unlock Eden you must defeat Mum’s heart. You can only play as Eden once then you can’t play as him again because you need Eden tokens. To get Eden tokens you need to defeat Mum’s heart, every time you kill Mum’s heart you get 1 Eden token. Eden starts with 2 random items.

Characters in Afterbirth, Afterbirth plus and Repentance.
There are a few characters added in each DLC. With new character and different functions for each one. here is how to unlock them.
Afterbirth
To unlock Lilith you need to beat greed mode as Azazel. Greed mode is a new mode in Afterbirth and can be played when you first get the DLC. You want a good run of Greed mode as Azazel and you will then unlock Lilith.
To unlock The keeper you need to donate 1000 coins to the greed machine. This a hard and frustrating thing to do as you can only donate 99 coins maximum each run and it would take 11 runs with the user getting 99 coins each time to donate 1000. But the problem is that the greed donation machine has a chance to jam whenever you put a coin into it. The keeper is hard to unlock.
Afterbirth plus
To unlock Apollyon you need to defeat Mega Satan. To do this you need to get both key pieces from the angel statues after blowing them up in the angel room. You can’t blow the statues up until you beat the Lamb or ??? once. Mega Satan is a hard to defeat boss.

Repentance
To unlock Bethany you need to defeat It lives with Lazarus without dying at all throughout the run. If you get a good run this isn’t that hard.
To unlock Jacob and Esau you need to defeat Mother.
